Directed and choreographed by Strictly Come Dancing judge Craig Revel Horwood, the musical stops off in cities including Birmingham, Malvern, Cambridge, Manchester, Liverpool, Glasgow, Sunderland, Leicester, Belfast and Bristol from July.
Based on the smash hit movie starring Whoopi Goldberg in the lead role along with Maggie Smith as a Mother Superior, Sister Act follows showgirl Deloris Van Cartier who witnesses a murder and hides out in protective custody at a convent.
Disguised as a nun and under the close watch of the Mother Superior, Deloris helps her fellow sisters in the choir find their voices.
Alexandra Burke, who has been touring in the lead role of Rachel in a UK tour of The Bodyguard, said: "I'm over the moon to be given the opportunity to play the wonderful Deloris in Sister Act.

The singer originally rose to fame after winning the fifth series of The X Factor and went on to become a pop star supporting Beyonce in concert. Her debut number one single, Hallelujah, sold over one million copies in the UK, a first for a British female soloist, and she went on to release singles Bad Boys and Start Without You.
This all new production of Sister Act features original music by the eight-time Oscar and Tony award-winner Alan Menken, who has created for the stage version of Disney's Beauty and the Beast as well as for films Tangled and Enchanted.

There's also another Tony award winner behind the scenes in the shape of Sarah Travis, who is responsible for musical supervision and arrangements for the show.
Craig Revel Horwood said: "I am truly thrilled that Alexandra is going to be our leading lady in this all new production of Sister Act. Alexandra is such a fantastic singer and actress that I know she is going to be amazing. I couldn't think of anyone better to play the role of the smart and funny Deloris."
Alexandra Burke will not be performing in any of the matinee performances on the tour.
