An initial ten-player field will compete at 16 venues across the country over four months in a bid to clinch the prestigious title, which was won by Gary Anderson last year.
Darts fans are getting braced to bring their 180 posters and fancy dress costumes to the lively session.
PDC Chairman Barry Hearn said: "The Betway Premier League has been a phenomenon in British sport for the past decade in taking the top player across the UK and into Ireland, with over 100,000 fans enjoying this year's tournament live across 16 great venues."
The opening nine Thursday nights see the players face each other once in the league phase, with the bottom two players from the league table being eliminated following Judgment Night.
The remaining eight players face each other once more over the remaining six league nights, one of which being hosted by the Barclaycard Arena in April.
The top four then progress to the play-offs in London, where the semi-finals and final will see the tournament's champion crowned.
The 2016 Betway Premier League will feature the top four players from the PDC Order of Merit following the completion of the 2015-2016 World Championship, with a further six players being invited to compete in the event.
